BCR::ABL1 Multiplex RT-PCR

Diagnostic Use

RT-PCR for detection of BCR::ABL1 transcripts commonly associated with the presence of a Philadelphia chromosome i.e. P190 (e1a2), P210 (e13a2, e14a2) and P230 (e19a2). The test can also detect some atypical BCR::ABL1 transcripts and these will then be characterised by Sanger sequencing.

Additional Specimen Information

12 ml peripheral blood in EDTA, 1-3 ml of bone marrow in EDTA or RPMI (anticoagulant).
Transport to the laboratory at ambient temperature within 48 hours of collection to ensure high-quality RNA.

Interpretation

Results from serial cell line dilutions determined a limit of detection of approximately 1 abnormal cell in 10,000 cells.
In the case of a negative result the RNA is confirmed to be adequate for analysis by amplification of a reference control gene.
A negative result does not exclude the presence of abnormal cells present at a level less than 1 in 10,000, or rare BCR::ABL1 transcripts.
This assay should only be used for determining presence and type of BCR::ABL1 transcripts. It is not a quantitative assay and should not be used for MRD purposes.
