Method

This method is adapted from that developed by Chomczynski and Sacchi and uses TRIZOL-LS reagent for the isolation of RNA from biological samples. WBC or tissue are homogenised in TRIZOL-LS, a monophasic solution of phenol and guanidine isothiocyanate. Addition of chloroform and centrifugation separates the organic and aqueous phases, and the RNA is recovered from the aqueous phase by isopropanol precipitation.

Stabilisation of RNA prior to storage or molecular testing.

Aliquot Instructions5-20 mL peripheral blood in EDTA. 1-3 mL bone marrow. 0.5 g tissue.
Aliquot Transport to CHLUrgent or overnight courier at room temperature or 4C. Samples to be received within 24 hours of collection. Dry ice transport for frozen tissues.
